The study was designed to test whether the effects of zolpidem and alcohol are simply additive or reflect potentiation. Skills performance impairment after acute doses of zolpidem (a short-acting, nonbenzodiazepine hypnotic), alone and with alcohol, was evaluated in 24 subjects.

In general, additive effects of alcohol were detected with zolpidem 10 mg but not with zolpidem 15 mg.

On day 6, 20 mg zolpidem was administered orally. In a randomized crossover study with two phases and a washout of 4 weeks, eight young healthy female volunteers took either 600 mg rifampin or placebo once a day for 5 days. Plasma concentrations and effects of zolpidem were measured up to 10 hours.

Rifampin (INN, rifampicin) is a potent inducer of many cytochrome P450 enzymes, and it greatly reduces the plasma concentrations and effects of, for example, midazolam, triazolam, and zopiclone. Zolpidem is a short-acting imidazopyridine hypnotic drug that is metabolized mainly by CYP3A4. In this study, the effects of rifampin on the pharmacokinetics and pharmacodynamics of zolpidem were studied.

It is likely that zolpidem also shows a reduced hypnotic effect when used concomitantly with other potent inducers of CYP3A4, such as phenytoin and carbamazepine. The effects of zolpidem are considerably reduced by rifampin because of enhanced metabolism of zolpidem.

The total area under the plasma zolpidem concentration-time curve after administration of rifampin was 27% of that after administration of placebo (336 +/- 67 versus 1202 +/- 157 ng.hr/ml [mean value +/- SEM; p < 0.01]). Rifampin decreased the peak plasma concentration of zolpidem by 58%, that is, from 293 +/- 61 to 117 +/- 25 ng/ml (p < 0.01) and the elimination half-life from 2.5 +/- 0.2 to 1.6 +/- 0.1 hours (p < 0.01). A significant (p < 0.05) reduction in the effects of zolpidem was seen in all six pharmacodynamic tests after rifampin pretreatment.

For each assessment, the maximal effect for both hypnotic drugs occurred 1.5 hour after intake. The effects of both hypnotic agents were short lasting, and no alterations were found 6 and 8 hours, respectively, after drug intake. Zolpidem is a rapid-onset, short-duration imidazopyridine hypnotic drug and is specific agonist of the omega-1 (BZD1) receptors. The aims of this study were to assess the effects of Zolpidem on memory (the main objective), psychomotor performances, and postural sway (secondary objectives) in 18 healthy subjects and to compare them with those of triazolam and placebo. No clinically relevant differences were found between Zolpidem and triazolam for memory, psychomotor performance, postural sway, or adverse effects. Its hypnotic characteristics resemble those of triazolam. Both drugs decreased psychomotor performance, impaired memory, and increased postural sway. Short-and long-term memory (paired words associate and pictures test), psychomotor performances (critical flicker fusion frequency, choice reaction time, digit symbol substitution test), and postural sway were evaluated before and 1.5, 4, 6, and 8 h after the administration of a single dose of Zolpidem (10 mg), triazolam (0.25 mg), and placebo. It may be concluded that Zolpidem, like triazolam, impairs short- and long-term memory, psychomotor performances, and postural sway and that these effects are of short duration.

A double blind, randomized, placebo-controlled study will be conducted in cardiac ICU patients who had been diagnosed with acute coronary.

Recent research has shown that SD, even in the short-term, may be related to echo and electrocardiographic changes that may potentially be predictors of cardiac arrhythmias. The environment of an Intensive Care Unit (ICU) is notoriously inhospitable to patients who experience a period of sleep deprivation (SD).

The objective is to evaluate the effects of early treatment with zolpidem controlled release (CR) compared to a placebo on clinical and polysomnographic parameters for patients in a cardiac ICU who had recently been diagnosed with acute coronary syndrome.

The study hypothesis is that sleeping better can improve the heart recovery in patients with a diagnosis of acute coronary syndrome.

Patients will undergo overnight full polysomnography on the first night in the ICU and will complete a sleep diary with a visual analogue scale to evaluate sleep quality in the morning after the first 3 nights of hospitalization. Inclusion Criteria: Exclusion Criteria:. A double blind, randomized, placebo-controlled study will be conducted in cardiac ICU patients who had been diagnosed with acute coronary syndrome. The results of the routine ICU laboratory tests including the serum levels of cardiac enzymes [troponin T and creatine kinase MB (CK-MB)] will be collected preceding the first dose of the drug/placebo, and then daily thereafter. The patients in group A will receive placebo and patients in group B will receive zolpidem CR 12.5 mg from the first night of hospitalization until their discharge.

It reduced sleep latency, increased total sleep time and decreased the number of awakenings during all recorded nights. Effects of zolpidem, a short acting non benzodiazepine hypnotic, have been studied in eight female poor sleepers, aged 25 to 54 years, documented during two consecutive adaptation nights. Subjects were recorded according to a 22 day single blind study. Zolpidem did not increase specifically stage 2 to the detriment of stages 3 and 4 but it restored them during the first nights of administration. Parameters of sleep continuity, sleep architecture and cyclical structure of sleep were studied. Placebo was given orally at bedtime on nights 2-4, zolpidem on nights 5-20 and finally placebo on nights 21-22. Polygraphic recordings (conventional analysis) were performed on nights 1-6; 12, 13; 19-22. During the first post-drug night a rebound insomnia was observed in two subjects only. Zolpidem 10 mg showed a hypnotic effect on poor sleepers. Finally, zolpidem had no effect on REM sleep temporal distribution. Study of the rapid eye movements-non rapid eye movements (REM-NREM) sleep cycles structure showed that the increase of stages 3 and 4 occurred only during the first part of the night.
